The text is evaluated at activation time.
When the first is discarded, the text of the second is not re-evaluated.
This is a constant in this game mechanism.

"You gain" is granted only if you don't have it, this is true through all cards.

You can find this weird or disturbing or not friendly, but this is a fact, and I doubt this will be changed soon.

So, if Adhikara had chain thanks to it being turn two... and played the big means, then was active again turn three (fog hydra setup)... they wouldn't have chain?

You know, it's still a bug if the mechanics are designed poorly... just a more meta one.

When you play the first amulet, it checks if you have the class. Check fails, you gain the class for as long as this amulet is in play.

When you play the second amulet, it again checks if you have the class. This time it passes so you gain the secondary effect for this amulet. On following turns there are no additional checks, so when the first amulet is removed, it does not change the secondary effect on the second amulet.

Simply said: A (permanent) card's effect is only checked when played (unless otherwise stated, like "and at the start of the fight") no matter how the trigger's change

Funnily enough, that's wrong.

A permanent cards defense bonus is reevaluated at the start of every turn, and can be cut off by a from behind with better initiative...

...so, if THAT text is reevaluated... why isn't the priest granting text?

never saw a from behind negating the effect of an item played before, like prestigious helment, etc.
